It's rare and very difficult to find, but "The Last Polka" with John Candy and Eugene Levy is the funniest movie I've ever seen.

I'll add The Pink Panther Strikes Again (that's the one where Dreyfus tries to destroy the world). And Stripes starring Bill Murray is one I always try to catch. Nobody mentioned Mrs. Doubtfire?

Trading Places---I just rewatched this for the first time in years and it's as funny as ever. The entire cast is great but Eddie Murphy is a force of nature. Plus can't beat Ralph Bellamy and Don Ameche, just an inspired bit of casting.
Tootsie--also rewatched recently. Had forgotten how good a film it is. Bill Murray steals the show as Dustin Hoffman's roommate.

Great movies, and have seen almost all of them, and mostly agree they each belong on a 'best of' list.
One I don't think has been mentioned yet, and that is definitely not the most funny ever, but was still plenty funny and also strangely endearing and quite witty at the same time, was "Tin Men" (Danny Devito/Richard Dreyfus/Barbara Hershey). Also visually stunning (the smorgasbord scene, Liberty Valance scene, and throwing clothes out into the yard come to mind).
When I had that on VHS, I think I wore the poor thing out.
Worth a view just for either the opening sequence of Cadillacs, or for the bar scene with Fine Young Cannibals providing background music.

Chevy Chase's best role. His dry delivery and demeanor in that movie (and its underrated sequel, Fletch Lives) are outstanding. I still find new funny things in them despite repeated viewings (probably over 100 for Fletch).

Jerry Lewis is superb in King of Comedy, playing a thinly-disguised Johnny Carson, paradoxically somewhat of a straight man for De Niro. Martin Scorsese was the director. A few of the scenes are among the best in all of comedy films, IMHO.

Tin Man not shown much anymore but I always watch it no matter at what point I find the movie on Cable.
my favorite part is when they scam the home owner with a phony Life magazine photo shoot and their before & after aluminum siding routine
and Barbara Hershey was at her peak gorgeous self - hard to believe she would be married to the Danny DeVito character

All of my favourites have already been listed except for one unless I missed it. I skimmed over the thread pretty quickly so I might not have caught it but O Brother, Where art Thou is near the top of my list and makes me laugh out loud every time I see it. Each time I watch I always catch nuances that I missed in the previous times.
